Abstract
Long-term live-cell imaging is a technique for the study of dynamic biological processes with high temporal and spatial resolution. This application note presents the benefits of using the X-Light V3 spinning disk confocal system from CrestOptics for extended time-lapse imaging of live samples. Thanks to its high acquisition speed, reduced phototoxicity, and excellent optical sectioning, the X-Light V3 is particularly well suited for advanced analysis in the field of Cell Biology. We describe two use cases: analysis of cell division dynamics in HeLa cells and real-time monitoring of autophagy using the mRFP-GFP-LC3 reporter. Our results demonstrate that the X-Light V3 enables long-term imaging with minimal photobleaching and high data fidelity, allowing robust quantitative analysis across hours of acquisition.
